چکیده

HE activity of xanthine dehydrogenase (XDH) in Drosophila melanogaster is affected by mutations at three loci: ma4 at locus 64ion the X chromosome, ry at locus 52 ion the third chromosome, and lxd at locus 33 ? on the third chromosome. See GLASSMAN (1965) for a discussion of these loci. The ma4 and the ry mutants are deficient in XDH while lzd flies have 20 to 25% of the XDH activity of wild type. To date, there has been no direct evidence that any of these loci is a structural gene for XDH. This paper presents data on electrophoretic variants of XDH due to changes at the ry+ locus.

Genetic limits of the xanthine dehydrogenase structural element within the rosy locus in Drosophila melanogaster.

Experiments are described that provide an opportunity to estimate the genetic limits of the structural (amino acid coding) portion of the rosy locus (3:52.0) in Drosophila melanogaster, which controls the enzyme, xanthine dehydrogenase (XDH). This is accomplished by mapping experiments which localize sites responsible for electrophoretic variation in the enzyme on the known genetic map of null-...

Hidden electrophoretic variation at the xanthine dehydrogenase locus in a natural population of Drosophila melanogaster.

Sixty-two isochromosomal lines of D. melanogaster were screened for cryptic electrophoretic variation at the xanthine dehydrogenase (XDH) locus. Sequential polyacrylamide vertical slab gel electrophoresis was performed using four electrophoretic criteria. A total of 15 classes of electromorphs were revealed. D. melanogaster appears to exhibit as much polymorphism at this locus as other extensiv...

Post-Translational Modification as a Potential Explanation of High Levels of Enzyme Polymorphism: Xanthine Dehydrogenase and Aldehyde Oxidase in DROSOPHILA MELANOGASTER.

Xanthine dehydrogenase (XDH) and aldehyde oxidase (AO) in Drosophila melanogaster require for their activity the action of another unlinked locus, maroon-like (mal). While the XDH and AO loci are on chromosome 3, mal maps to the X chromosome. Although functional mal gene product is required for XDH and AO activity, it is possible to examine the effects of mutant mal alleles in those cases when ...

Intracistronic mapping of electrophoretic sites in Drosophila melanogaster: fidelity of information transfer by gene conversion.

A convenient method is described for the intracistronic mapping of genetic sites responsible for electrophoretic variation of a specific protein in Drosophila melanogaster. A number of wild-type isoalleles of the rosy locus have been isolated which are associated with the production of electrophoretically distinguishable xanthine dehydrogenases. Large-scale recombination experiments were carrie...

Organization of the rosy locus in Drosophila melanogaster: evidence for a control element adjacent to the xanthine dehydrogenase structural element.

From a collection of electrophoretic variants of XDH obtained from laboratory strains and natural populations, a stock was isolated that was associated with much greater than normal levels of XDH activity. Preliminary recombination experiments demonstrated that this character maps to the rosy locus. While a series of observations failed to relate this phenotype to alteration in the structure of...
